The Relationship between T869C Polymorphism of Transforming Growth Factor-¥â1(TGF-¥â1) Gene and Change in Bone Mass after Hormone Replacement Therapy in Postmenopausal Korean Women

Abstract
Objectives: To investigate the relationship between the T869C polymorphism of transforming growth factor-¥â1(TGF-¥â1) gene, and change in bone mineral density(BMD) after hormone replacement therapy(HRT) in postmenopausal Korean women.

Methods: The T869C polymorphism of TGF-¥â1 gene was analyzed by allele specific polymerase chain reaction(PCR) and electrophoresis in 302 postmenopausal Korean women receiving sequential HRT for 1 year. Serum bone alkaline phosphatase(BAP), CrossLaps(CTX), and TGF-¥â1 were measured by immunoassay, and enzyme linked immunosorbent assay respectively. BMD at the lumbar spine and proximal femur was determined by dual energy X-ray absorptiometry.

Results: The TGF-¥â1 genotypes were not related with annual percent change of BMD at lumbar spine and proximal femur after HRT, and were not distributed differently between HRT-responders and HRT-nonresponders(women who lose more than 3% of bone mass per year). There were no associations between serum TGF levels or their 6 month percentage changes after HRT and basal BMD or annual changes in BMD after HRT. The change in serum TGF-¥â1 level did not correlate with change in serum levels of bone turnover markers after HRT.

Conclusion: The T869C polymorphism of TGF-¥â1 gene is not associated with change in BMD after HRT in postmenopausal Korean women,
